Materials:
- Stir Bar
- Small Beaker (20 mL)
- Spatula
- Parafilm
- Graduated Cylinder
- Fine-Balance Scale
- Stir Plate
- 20 ml Milk Bottle Wrapped in Foil (sterile)
- Filter Sterilize Equipment
- 10 mL Syringe
- 0.22 uM Filter
- Fume Hood or Bunsen Burner
Protocol:
- Take out Kanamycin Sulfate from the desiccator
- Measure out 0.5 g into the beaker on the fine-balance scale
- Add 5 mL of distilled water to the beaker and a small stir bar, put parafilm on top to prevent spills. Then place the beaker on a stir plate to dissolve.
- Once dissolved, measure the solution with a small graduated cylinder and add distilled water until the total volume is 10 mL
- Filter sterilize solution using the syringe and filter and working near a flame or in the biosafety cabinet. Take off the plunger of the syringe and carefully add the filter to the bottom of the syringe. Pour the solution into the top of the syringe (where the plunger was). Move the syringe so the filter is now over the milk bottle. Add the plunger back on top and slowly push the plunger until all the solution has been filtered into the milk bottle.
- Store at 4°C.
