Media used for double cross over screening for pkoV allelic replacement.

| Total volume of media | 400 mL | 800 mL | 1800 mL |
| Tryptone | 4 g | 8 g | 18 g |
| Yeast Extract | 2 g | 4 g | 9 g |
| Distilled water | 400 mL | 800 mL | 1800 mL |
- Prepare media in desired volumes.
- Carefully add a stir bar to bottle and mix ingredients. Once mixed, remove stir bar using magnetic stick.
- Autoclave on a liquid load for at least 45 minutes.
